Passive. Your missiles have a chance to pass through enemies that they hit.
Pierce is a level 30 Amazon passive (Passive and Magic tree, requires Penetrate; it unlocks nothing). Every missile the Amazon fires or throws that lands has a chance to keep flying and hit what stands behind. The numbers that decide it are the chance, 85% at level 20, and the limit of four pierces per missile.

dm12, the diminishing-returns calc that runs from Param1 (the minimum, 10) toward Param2 (the maximum, 100). The Basin's page writes it out as min(10 + [90 × [(110 × slvl) ÷ (slvl + 6)] ÷ 100], 100), and the table's values follow it: 23% at level 1, 55% at 5, 71% at 10, 80% at 15, 85% at 20, 89% at 25, 91% at 30. The Basin's table reaches 100% only at level 60.The calc reads lvl, so +skills from items raise the chance; the passive is applied through passivestat1, skill_pierce, the same stat the Piercing Attack affix on items sets.
Pierce is a passive state (pierce) with no cast and no radius. By the Basin's page it applies to arrows, bolts, javelins, throwing axes and throwing knives, and the roll is made whenever the missile's hit check succeeds or is skipped by an attack that always hits. A missile that pierces flies on and makes a fresh Chance to Hit roll on the next target, with its full damage and modifiers; the Basin's page sets the limit at four pierces, so one missile can hit up to five targets. If a target blocks the missile (Blocking), the damage and modifiers are not applied to that target, but the block does not stop the missile piercing on.
The Basin's page warns that when the hit check fails but the pierce roll succeeds, the missile may appear to pass through and even play its effects on the next target, but it does not really pierce: nothing behind the missed target takes damage or any modifier. A missile pierces only from a hit.
The chance stacks additively with Piercing Attack from items, by the Basin's page: the Amazon's passive and the item affix are the same stat.
No synergies: the row has no SymPerCalc column, the skilldesc row has no dsc3 line, and no skill reads Pierce's points. The chance reads lvl, so +skills from items count in full; it is the last skill in the tree, so nothing is gated behind it.
A player's hit on another player is cut to 17% of its value (difficultylevels.txt PlayerDamagePercentVSPlayer) before anything else, see Player versus Player; Pierce adds no damage of its own, so each hit a pierced missile lands on a player is cut the same way. The Basin's page says nothing more about this skill against players.
