Stamina is the bar that drains while you run. When it is empty you drop to walking until it refills. Maximum Stamina makes the bar longer; it does nothing else.
Every class starts with a base amount and gains more per level and per point of Vitality (charstats.txt stamina, StaminaPerLevel, StaminaPerVitality; the last two are stored in quarter points, converted here):

The Assassin also drains stamina more slowly while running (charstats.txt RunDrain 15, against 20 for every other class).
Templar's Might and Aldur's Advance add a lot of it. A stamina bar that never empties is usually reached sooner with Slower Stamina Drain or Heal Stamina Plus.
